I Boofed 800mg of Melatonin: Here's My Story
800 mg of melatonin can truly make you question your existence.
I remember lying naked on my bathroom floor, with two bullet-shaped molds of melatonin and coconut cream to my right and a YouTube video called "How to Do a Suppository" playing on my left.
What led me to this situation, both literally and figuratively, was the idea of a supraphysiologic dose. A dose so substantial it could significantly rewire the health of body and mind for extraordinary performance.
You might be shocked by the amount I took. You might think you'd see demons at that dose. I did too. But after careful consideration of the science, I thought, let’s ride.
And here we are. So, let's dive deep into my experience with 800 mg of melatonin.
BEFORE THE DOSE
I became intrigued by this concept during my junior year of college.
I was deep into biohacking and absolutely fascinated by all the quirky elements in the scene. From red-light glasses , kava infused beverages and weird EMF beanies. I’m a biohacker addict. Take me to a BA meeting.
One day I was listening to John Lieurances podcast on melatonin.
This guy absolutely loves the stuff. He loves it so much he call is the “magic bullet.”
He began the show talking about the misconceptions surrounding high doses of melatonin. He also mentioned that many doctors are misinformed about melatonin and are missing out on its therapeutic benefit.
The reason I became interested in melatonin was because of its role in autoimmunity:
Melatonin is considered a pleiotropic molecule, with both pro-inflammatory and anti-inflammatory properties. It is immunomodulatory, meaning it reduces excessive immune function in inflammatory conditions while improving immune conditions in immunocompromised individuals. So, you could say melatonin is truly adaptogenic, ready to meet whatever needs it calls for. - Dr. John Lieurance
For someone who has a susceptibility to CIRS (chronic inflammatory response syndrome), I was looking for something to help with that, and out of all the biohacks that were available, I think this was the one.
I was basically convinced, but I wanted to know one last thing before trying this.
Will I stop producing my own melatonin if I do this?
Short answer: no. There is no negative feedback loop with melatonin. Unlike testosterone therapy, which signals the body to stop producing endogenous testosterone, exogenous melatonin does not inhibit our natural production.
Game on.
I searched for high-dose melatonin all throughout the land. Unfortunately, CVS doesn’t sell the potent stuff, so I had to visit a holistic medicine clinic. You can find one by searching for a functional medicine clinic, regenerative health, or holistic health clinics near you and see if they got the goods.
Fast forward, and I’m at my favorite biohacking clinic.
With rows of multivitamins, probiotics, red-light therapy, you could say I'm in heaven.
“Right this way, Jack.”
The nurse reaches into the fridge and pulls out a silver-laminated bag with the words “silver bullet” on the cover. Beneath the title were the words, melatonin, 400 mg.
You’ve done this before, right? she asked playfully yet informatively.
Yeah, for sure. High-dose melatonin is going to be a breeze, I thought.
THE DOSE
Fast forward to my bathroom tile, and I'm still astonished that suppository meant up the butt.
There was no time to back out. I set aside my toxic masculinity and lay my cold backside on the tile.
I took a deep breath and inserted the bullet-shaped mold. It felt bizarre, yet not painful. It was like taking a reverse dump.
Once you push it in all the way, it vanishes into your colon. It’s as if it got swallowed. Then it was done. Some doctors recommend elevating your legs on the wall for a couple of minutes afterwards or hanging upside down on a bar. I put my legs up for that ultimate absorption brah.
The next morning, I woke up feeling as if I was on Xanax. I stood up and almost fell over. My mind was clear, and my body felt rejuvenated though. It was the type of morning when you wake up with an immediate smile. Something beautiful occurred in my body that night, and I wanted to explore more of that.
I continued this routine for the week. I ended up taking two suppositories to create an 800 mg dose. The supraphysiologic dose. That felt even weirder. Sleep is strange at such a high dose; I hardly remember it, and you could sleep for 8-10 hours yet feel like only a minute passed. Dreams were vivid; you definitely don’t wake up. I’m pretty sure my Oura ring scores were soaring during this period, approaching almost 100 every night.
Before we continue, why did I choose to take melatonin rectally? Why not just consume 100 capsules?
It all relates to blood plasma volume.
Blood plasma volume (BPV) refers to the amount of a substance that enters your bloodstream. The higher the BPV, the greater the quantity and more effective the compound. Suppositories provide the highest BPV of any delivery mechanism, even IV.
Post Experiment
It's been a couple of years since my infamous self-experiment.
I love melatonin and take it regularly. When traveling, I use it to reset my circadian rhythm and will take it nightly. If I’m at home and can’t sleep, I use it too. I typically take anywhere from 50-100 mg at home. After traveling, I now take 200 mg. Maybe I’ll try 400mg or 800mg again, but right now, there's no need.
Most days, I take little melatonin. I feel better when I don’t take it for a while and then do a dose. If I use it every day for weeks, I feel like I’m clogging my sleep systems, so I prefer a more natural approach. But it's one of the tools in my biohacking arsenal 100%.
Do I still take suppositories?
No, I now use troches, which absorb decently and aren’t as tedious as a suppository. I take one by Mitozen that also contains methylene blue and probiotics.
Most nights, I stick to sleep breakthrough and magnesium breakthrough by BiOptimizers, which provide all the precursors to melatonin and a formulation for great sleep.
Where should you get melatonin?
Be cautious with it; don’t buy random melatonin from CVS and expect quality. Some are improperly dosed and filled with nonsense. You should purchase from reputable companies that offer quality delivery forms and products. My favorite melatonin company is Mitozen.
Overall summary
Melatonin can be a valuable tool if dealing with insomnia, circadian rhythm misalignment, or illness, but it should only be used as a tool. It shouldn't replace poor sleep habits or diet. Check out this sleep guide if you want to build a great sleep environment.
Summary:
Melatonin has significant medicinal properties worth exploring. I learned about this in Dr. John Lieurance's book on melatonin.
Avoid subpar melatonin. Opt for quality products from trustworthy companies.
Most nights, I prefer taking melatonin precursors and allowing my natural melatonin production; it feels pleasant and doesn’t leave me bogged down like some melatonin does.
Here is the sleep breakthrough and magnesium supplement I take every night.
That’s all today fam. Hope yall enjoyed. Happy 2025!!!!
I’m grateful to my partners who create amazing health & wellness products.
Disclaimer. I am not a doctor. This is just some random stuff I learned on the internet. Pretend it’s a story. Ok bye.

Very interesting, Jack. We have never heard of macrodosing melatonin before stumbling across your post. Did you journal your dreams?
That sounds like it’s quite the experience. And no down sides?
